I've been stubborn about keeping my favorite saddle for the last 4 months. I have a Liquigas Supersix with a special edition Danilo DiLuca "Killer" F'iz:ik Arione saddle - absolutely love the look. Unfortunately, I've had constant numbness in my member, even on my off days. I desperately tried everything to get comfortable - saddle forward, tipped up/down, seatpost up/down. I'd get some improvement in comfort (with a slightly tipped up, forward saddle with seatpost down about a cm), but no relief. Finally, I found a saddle that almost rivalled the look of the Killer saddle in the World Champion Toupe Team saddle. I sized my pelvis and ordered a 143 size from one of my LBS. I kept the everything the same and replaced the saddle - so far, numbness subsided and no problems. I will say that it isn't the most comfortable saddle in the world with no padding, but I don't find myself squirming while I ride. I guess it's no coincidence that I read many great reviews similar to mine about the Toupes.

As a follow-up, I've started to notice that the Toupe Team is getting a little more uncomfortable the more time I spend in it. Luckily, the numbness continues to subside. However, my ischial tuberosities (sitz bones) are taking a beating. I think it may be that the saddle isn't aligned correctly - I'll try moving it backward, effectively narrowing the saddle. It seems a tad better on my backside when I sit forward, but I'd never call it comfortable. Somehow, the Arione and the Toupe seem to offer different things - the Arione doesn't bother my bones, but makes me numb, while the Toupe is the converse.

Anyone have any suggestions? I'm considering trying a Selle Italia Flight Gel Flow next.

Well, don't move the saddle back. Saddle position places your legs correctly with respect to the BB.

Some people have found that the Toupe takes a bit of time to break in, or so they say. My Toupe was comfy from the start and has stayed that way for the last year.

1 day isn't that long of a trial. Also, saddles don't have one universal correct position, so with your change to a Toupe, it's likely, or at least possible, that you need to change the angle on the saddle to find its sweetspot.
